Protein-RNA Interactions and Post-Transcriptional Modulations

The Peng Wu Group is developing novel small-molecule-based chemical modulators to manipulate the pervasive protein–RNA interactions, which impact almost all aspects of RNA biology and essential cellular events. Deregulations of the complex protein-RNA interaction network and the resulting post-transcriptional modifications are associated with various human diseases, while very limited chemical modalities and modulation approaches are available. The Wu group develops platform strategies based on small molecules to tackle the challenging interactions between RNA-binding proteins and disease-associated RNAs. The goals are to discover new chemical probes to study the biological networks contributing to the expression and function of nucleic acids, and to provide lead compounds for small-molecule-based therapeutics with understudied mechanisms of action at the post-transcriptional level.

The Wu Group also develops related scaffold-diverse synthetic strategies to access these biologically active small molecules, as well as strategies to synthetically construct new chemical modalities and bifunctional molecules modulating the degradation and modifications of RNAs and RNA-binding proteins.
